Evaluation of a Loop-Mediated Isothermal Amplification (LAMP) Method for the Detection of Salmonella spp. in Terms of Sensitivity and Applicability

Evaluation of a Loop-Mediated Isothermal Amplification (LAMP) Method for the Detection of Salmonella spp. in Terms of Sensitivity and Applicability

Review Data

Purpose and Significance of Study: The study involved the successful development of two LAMP assays with genomic DNA from pure bacterial cultures of Salmonella spp., and the invA gene and 16S-23S rRNA intergenic spacer region of Salmonella enterica as target sequences, and their specificity and sensitivity were evaluated. This report can provide a baseline for the application of LAMP for detection of food-borne pathogenic bacteria.

Fit with Scope of Journal: The manuscript is of very high interest for the journal Journal of Food Nutrition and Metabolism.

The Introduction places the study in clear context, the Methods are Results aptly convey the details of the study, and the Discussion adequately explains the implications of the findings.

o   The manuscript contains a few errors (already corrected in the galley proof, except the highlighted one, which requires the author’s attention), which are listed below –

·       In the 2nd sentence of the Abstract, it is claimed that “more than 20 detection methods have been established in Salmonella spp. using loop-mediated isothermal amplification (LAMP) or reverse transcription loop-mediated isothermal amplification (RT-LAMP)”; however, the actual number seems to be much more (please refer to this article)

·       In the last sentence of the 2nd paragraph of the Discussion, the part “The mechanisms of…are clear” appears incomplete and therefore, does not make clear sense; hence, it must be rephrased or the context must be added.

·       Throughout the text, “et al” must be followed by “.”.
