Purpose and Significance of Study: This study investigated the anti-tumoral role of the small molecule analog and orexin antagonist, almorexant, in colon cancer. The anti-tumoral effect of almorexant was possibly mediated by orexin receptor 1. The finding poses almorexant as a putative candidate for the treatment of colorectal cancer, one of the most prevalent of cancers.

 

Fit with Scope of Journal: The manuscript is of very high interest for the journal European Journal of Molecular Cancer.

 

In the discussion, the authors have elaborated the anti-tumoral effect of almorexant on the HT-29 colon cancer cell line, compared that to the effect of orexin, discussed the possible pathway of action, and also its effect in a preclinical model, all of which contributes to the potential therapeutic implications of almorexant in colorectal cancer.

 

The manuscript contains a few minor errors (already corrected in the galley proof), which are listed below

 

·       Using incorrect tense, e.g., “led” after “prostaglandins” in the 10th sentence of the Introduction must be replaced with “lead”, “produced” in the 12th sentence of the same paragraph must be replaced with “produces”, and so on.

·       Using “to” and “of” instead of “on” after “depend” and “with” after “associate” on multiple occasions.

·       In the 7th sentence of Almorexant induced a mitochondrial apoptosis in colonic cancer cell line HT-29 in the Results and Discussion, “discriminate” must be replaced with “distinguish”.
