MMP-9 Shedding Releases Truncated CD73 from Astrocytes

Purpose and Significance of Study: This study attempted to investigate the mechanism leading to the loss of membrane CD73 in astrocytes. The results indicate that under active inflammatory condition, MMP-9 releases CD73 from astrocytes. The block of CD73 shedding in astrocytes by the addition of MMP-9 inhibitor could significantly decrease the activation of interacted effector T cells.

Fit with Scope of Journal: The manuscript is of very high interest for the journal Neurology and Neurobiology.

The Discussion presents a thorough analysis of the results of the study and aptly lays down the implications.

 

o   The manuscript contains some minor errors (already corrected in the galley proof, except the highlighted one, which requires the author’s attention) are listed below –

 

·       In many places throughout, “;” is used instead of “,”, e.g., after “CD73” in the 2^nd sentence of the Abstract, after “blocked” in the 6^th sentence of the Abstract, and so on.

·       Some words are misspelled, e.g., “form” must be replaced with “from” in the 1^st sentence of The interaction of isolated astrocytes and CD4 T cells under Materials and Methods, and “immnupopricipitated” must be replaced thrice with “immunoprecipitated” in C-terminal truncated CD73 was released into medium under Results.

·       In the Abstract, in the 1^st sentence, “expresses” must be replaced with “express”, and in the 4^th sentence, “of” before “Cd73” must be replaced with “in”.

·       In the 1^st sentence of the 1^st paragraph of the Introduction, “participate” must be replaced with “participates”.

·       In the 2^nd paragraph of the Introduction, in the 4^th sentence, “hindered” must be replaced with “hindering”; in the 5^th sentence, “understanding” must be replaced with “understand”, “of” before “local” must be replaced with “about”, and “finding out” must be replaced with “find”; in the last sentence, “Which” must be replaced with “This”, and “also” must be removed.

·       In the 3^rd sentence of The induction of EAE under Materials and Methods, “i.p.” must be replaced with “IP”.

·       In the 3^rd sentence of The detection of Cd73 mRNA expression in astrocytes in the Materials and Methods, “paralleled” must be replaced with “parallel”, and in the last sentence of Immunoprecipitation of CD73 in the Materials and Methods, “send” must be replaced with “sent”.

·       In the 3^rd sentence of the detection of active form of MMP-9 in the Materials and Methods, “stay” must be replaced with “and kept”.

·       In C-terminal truncated CD73 was released into medium under Results, in the 1^st sentence, “was” before “cultured” must be replaced with “were”, and in the 3^rd sentence, “was being” must be removed.

·       In the last sentence of the 1^st paragraph of the Discussion, “of promoting” must be replaced with “to promote”, and “significantly” must be replaced with “significant”.

·       In the 8^th sentence of the 3^rd paragraph of the Discussion, “accounted” must be replaced with “accounting”, and “lose” must be replaced with “loss”.

·       The last sentence of the last paragraph of the Discussion is incomplete; hence, it must be completed.
