Discussion & Conclusion

Q: Is it related to the results presented before? Do you consider them as coherent?

A: Good

Comments: The Discussion states that the targeted therapies have helped make great progress in the treatment of acute myeloid leukemia (AML). The Discussion illustrates that out of the 4 candidate genes identified integrin β1 (ITGB1) is the hub gene related to the bone microenvironment mediated poor prognosis in AML and compared to normal people, the expression of ITGB1 is significantly lower in AML patients. The Discussion establishes that the ITGB1 may be a key gene for evaluating the prognosis and conducting targeted therapy of AML patients. The study concludes that its findings will provide a theoretical basis for a deeper understanding of the occurrence and development of AML which will also give the follow-up animal experiments and clinical trials research directions.

Comments: Except the following errors have been detected:

1.     The 5th sentence under the subheading Analysis and Screening of Differentially Expressed Genes (DEGs) under Materials and Methods was not framed properly and should be changed to “Then, the R package “limma” was used to differentiate the…. of ATO.”

2.     The 2nd sentence under the subheading Analysis and Screening of Differentially Expressed Genes (DEGs) under Results was not framed properly and should be changed to “Performing statistical analysis on the GEO database, we…. respectively).”

3.     The words “reveal” and “perform” have been misspelled in the text part of the manuscript.

Further comments on the paper

Comments: The study aims at finding the hub gene of acute myeloid leukemia (AML) related to the bone marrow microenvironment mediated multidrug resistance (MDR). The study screened out the differentially expressed genes (DEGs) of AML cells to find potential key biomarker of AML related to the bone marrow microenvironment mediated poor prognosis, by using biological information technology and the analysis of high-throughput sequencing chip data. The study identified integrin β1 (ITGB1) as the hub gene that can be considered as the predictor of prognosis. At the molecular level, the occurrence and development of AML has been established to provide a potential guidance for targeted therapy. The study highlights that the patients with high expression of ITGB1 had significantly longer overall survival (OS) than those with low expression. The study concludes that ITGB1 is a hub gene related to the bone microenvironment mediated poor prognosis in AML and the expression of ITGB1 is negatively correlated with the prognosis of AML. Future studies are needed to further investigate ITGB1 as the potential marker for predicting and guiding the treatment of AML.
